The microbiological assay for vitamins has been a valuable analytical method because of its great sensitivity and ease of setting up. However, when applied by several investigators to the detection of the folic acid activity of human serum and whole blood, the results have not always been satisfactory. In many cases, this has been due to the fact that insufficient attention was paid to the basic design of the assay. In addition, the biological activity of the naturally occurring folates presents special problems. In this article, the various phases of the microbiological assays for folic acid activity have been considered and a detailed scheme, based on well-established scientific principles, presented.
