Albumin synthesis by the isolated perfused rat liver was measured by the (14C) carbonate method using perfusates of whole rat blood with and without the addition of 400 mg/100 ml alcohol. We found albumin synthesis significantly reduced by alcohol. The addition of a mixture of 11 essential amino acids to 10 times their normal plasma concentration restored albumin synthesis to normal in the presence of 400 mg alcohol. Omission of tryptophan from the amino acid mixture did not affect the ability of amino acids to restore albumin synthesis to normal. Correction of disordered NAD/NADH ratio, due to alcohol, did not reverse the effects of alcohol on albumin synthesis.
